Ku Proteins Join DNA Fragments as Shown by Atomic Force Microscopy1

چکیده
The binding of the Ku protein to DNA was investigated using the atomic force microscope@Ku was found to bind predominantly to the ends of double-stranded DNA. Experiments with plasmid DNA revealed that Ku does not bind to circular plasmids but does bind to plasmids that have been linearized by treatment with ionizing radiation. The binding ofKu to poly(dG-dC)poly(dG-dC) polynucleotides and to a 400-bp DNA EcoRI fragment resulted in a shift in the fragment size distribution to include longer fragments, with internally binding Ku. Furthermore, we observed images consistent with fragments joined together by Ku, showing an interacdon with two ends of DNA. These observations suggest that Ku may play a role in physically orienting DNA for ligation by binding the ends of adjacent DNA molecules.

متن کاملDNA looping by Ku and the DNA-dependent protein kinase.
The DNA-dependent protein kinase (DNA-PK) is required for DNA double-strand break (DSB) repair and immunoglobulin gene rearrangement and may play a role in the regulation of transcription. The DNA-PK holoenzyme is composed of three polypeptide subunits: the DNA binding Ku70/86 heterodimer and an approximately 460-kDa catalytic subunit (DNA-PKcs). DNA-PK has been hypothesized to assemble at DNA ...
متن کاملDNA studies using atomic force microscopy: capabilities for measurement of short DNA fragments
Short DNA fragments, resulting from ionizing radiation induced DNA double strand breaks (DSBs), or released from cells as a result of physiological processes and circulating in the blood stream, may play important roles in cellular function and potentially in disease diagnosis and early intervention. The size distribution of DNA fragments contribute to knowledge of underlining biological proces...

Unrepaired double-strand breaks (DSB) in the sugarphosphate backbone of the DNA molecule resulting from exposure of cells to ionising radiation are believed to be critical lesions that can lead to cell killing, mutation and carcinogenesis. Moreover, the pattern of DNA fragmentation is directly related to radiation track structure and, in the case of high-LET radiation, becomes determined by the...
متن کاملDNA fragmentation – dependence on the radiation quality
Ionizing radiation produces a plethora of lesions in DNA including strand breaks (single or double) and alteration to bases. Double-strand breaks (DSBs) leading to DNA fragmentation are considered to be the most critical lesions resulting in cell killing, cancerogenesis or mutations [1] High-LET radiation produces more complex and localized damage than X-rays due to induction of spatially corre...
